Anti-Trimeresurus albolabris venom IgY antibodies: preparation, purification and neutralization efficacy J. Venom. Anim. Toxins incl. Trop. Dis.
Duan,Hai-long; He,Qi-yi; Zhou,Bin; Wang,Wen-wen; Li,Bo; Zhang,Ying-zheng; Deng,Qiu-ping; Zhang,Ying-feng; Yu,Xiao-dong.
Abstract Background Snakebite incidence in southwestern China is mainly attributed to one of the several venomous snakes found in the country, the white-lipped green pit viper Trimeresurus albolabris. Since antivenom produced from horses may cause numerous clinical side effects, the present study was conducted aiming to develop an alternative antivenom antibody (immunoglobulin Y - IgY) from leghorn chickens. Methods IgY in egg yolk from white leghorn chicken previously injected with T. albolabris venom was extracted by water, precipitated by ammonium sulfate and purified by affinity chromatographic system. IgY was identified by SDS-PAGE, ELISA and Western blot, and its neutralizing assay was conducted on mice. Results Chickens injected multiple times...

Comparison of Mycoplasma gallisepticum Infection in Different Samples and Ages of Chicken Breeder Flocks Rev. Bras. Ciênc. Avic.
Demirbilek,SK; Ardicli,Ö; Carli,KT.
ABSTRACT This study aimed to compare method-based and newly developed sample-based methods for Mycoplasma gallisepticum (MG) detection in different samples of breeder flocks suffering from respiratory disease problems by using culture, real-time PCR (rPCR) and ELISA from chicks and embryonated eggs. Overall, 450 samples of 19-day-old chicken embryo’s trachea, 450 samples of 8-day-old chicken tracheal swabs and 900 blood samples of 20-, 27-, 34-, 40- and 46-week-old breeder chickens from 5 flocks were sampled for 26 weeks, and were all tested for MG by culture, MG-rPCR and MG-ELISA. Culturing assays and rPCR were applied to 450 mixture samples from 19-day-old chicken embryo’s trachea and 450 tracheal swab samples (each pooled into groups of 3) from...

Development of an enzyme-linked immunosorbent assay for quantification of Salmonella enteritidis-specific antibodies in egg yolk Rev. Bras. Ciênc. Avic.
Tayeb,IT; Nehme,P; Jaber,L; Barbour,EK.
The present study aims at developing an indirect ELISA to quantify yolk antibodies specific to all surface proteins of the invasive Salmonella Enteritidis (SE), which acquired the 1.8, 14.1, and ~ 50 Kb plasmids. An ELISA checkerboard was used in four different experiments to account for the different parameters included in the preliminary ELISA procedure, and consequently to maximize the difference in Optical Density (OD) values between control positive and negative yolk samples. The first experiment aimed at studying the impact of 5% Bovine Serum Albumin (BSA) dissolved in distilled water as a blocking reagent on a 28 µg/well SE antigen-coated plate, while applying the positive and negative control yolk samples to different concentrations of...

Dietary intake of broiler breeder hens during the laying period affects amino acid and fatty acid profiles in eggs R. Bras. Zootec.
Li,Feng; Yang,Yi; Yang,Xue; Shan,Mingxu; Gao,Xiang; Zhang,Yuanyuan; Hu,Jingwei; Shan,Anshan.
ABSTRACT The objective of this study was to evaluate the effects of maternal dietary intake on the amino acid and fatty acid contents in eggs during the laying period. An experiment with a 2×2 factorial design was conducted with two maternal dietary intake levels (100 and 75% of dietary intake recommended by the Chinese Ministry of Agriculture) and two broiler lines (fat and lean line). A total of 384 hens of fat line and 384 hens of lean line at 23 weeks of age were included in the experiment. Each line was randomly divided into two treatments (n = 192 in each treatment, with 12 replications), and each replication included 16 birds. The treatments were LN (lean line and normal maternal dietary intake, n = 192), LL (lean line and low maternal dietary...

Effects of carob ( Ceratonia siliqua ) pod byproduct on quail performance, egg characteristics, fatty acids, and cholesterol levels R. Bras. Zootec.
Calislar,Suleyman; Kaplan,Yusuf.
ABSTRACT This study was carried out to determine the effects of a carob (Ceratonia siliqua) byproduct (CB) supplement in diets for laying Japanese quail (Coturnix coturnix japonica) on performance and egg internal-external quality traits, fatty acid profile, and cholesterol content. A total of 225 female quail at 12 weeks of age were distributed into five treatment groups with three replications (15 birds in each replication). The following treatments were tested: 0% (control, no CB supplementation); 3% CB; 5% CB; 10% CB; and 15% CB in the diets. Feed and water were provided ad libitum to quail. The lighting program was 16 h light and 8 h dark. The experiment lasted 60 days. Treatments had no significant effects on final weight, feed intake, feed...

Elisa evaluation of the levels of antibodies against Infectious Bronchitis Virus in laying hens using egg yolk as substrate Rev. Bras. Ciênc. Avic.
Rauber,RH; Flôres,ML; Pereira,CE; Grigulo,M.
The present work was carried out to compare Infectious Bronchitis Virus (IBV) antibody titers in serum and egg yolk samples from laying hens. Sixty paired blood and egg samples were collected from laying hens of two farms. Serum samples were frozen, while egg yolk samples were diluted (1:500) before freezing. Serum and yolk samples were tested for the presence of IBV antibodies by indirect ELISA (commercial kit) and titers were compared by a correlation test (alpha=0.05). There was a high correlation (r=0.62) between the two kinds of samples, which means that titers of IBV antibodies in the egg yolk and in serum samples are quite the same. Considering that blood collection causes deep stress that leads to economic losses, and since eggs are collected daily...

Preparation and neutralization efficacy of IgY antibodies raised against Deinagkistrodon acutus venom J. Venom. Anim. Toxins incl. Trop. Dis.
Liu,Jinhua; He,Qiyi; Wang,Wenwen; Zhou,Bin; Li,Bo; Zhang,Yingfeng; Luo,Cong; Chen,Diancheng; Tang,Jia; Yu,Xiaodong.
Abstract Background The five-paced pit viper (Deinagkistrodon acutus), endemic to China and northern Vietnam, is responsible for most snakebites in the Chinese territory. Antivenom produced from horses is the main treatment for snakebites, but it may cause numerous clinical side effects and have other disadvantages involved in their production such as the welfare of animals. The present study was conducted aiming to develop an alternative antibody (IgY) from the egg yolk of leghorn chickens immunized with snake venom. Methods IgY from the egg yolk of white leghorn chickens previously immunized intramuscularly with D. acutus venom was extracted by water, precipitated by ammonium sulfate and purified by affinity chromatographic system. IgY was identified...
